Abstract Text: Calcineurin-responsive zinc finger 1 (Crz1) is a key target of the calcium (Ca2+)-signaling protein calcineurin, regulating growth, stress tolerance, and virulence in fungi. Our previous studies identified FOXG_05246 and FolCRZ1 as homologs of yeast CRZ1 in Fusarium oxysporum f. sp. lycopersici (Fol). Unlike FolCRZ1, FOXG_05246 lacks conserved calcineurin-binding motifs, suggesting it is not a calcineurin substrate. Notably, the FOXG_05246 mutant exhibited higher oxidative stress sensitivity and reduced virulence compared to the FolCRZ1 mutant, indicating its unique role in Fol’s cellular responses. Here, we investigated how FOXG_05246 mediate the oxidative stress response and virulence. Transcriptome analysis revealed 912 upregulated and 964 downregulated genes in the FOXG_05246 mutant compared to the wild type. Gene ontology and KEGG analysis showed that the differentially expressed genes were related to oxidoreductase activity, protein phosphorylation, DNA damage response, and protein degradation, while antioxidant enzyme gene expression remained unchanged, suggesting that FOXG_05246 may counteract oxidative stress indirectly by modulating redox balance, protein signaling, and cellular repair processes, thereby influencing virulence. Subcellular localization analysis using a FOXG_05246-GFP fusion strain showed translocation of FOXG_05246 from the cytoplasm to the nucleus under oxidative stress, further supporting its potential role as a transcription factor. This study identifies FOXG_05246 as a novel regulator of oxidative stress resistance for Fol pathogenicity.
